How qPCR preamplification kit supports low-input analysis

Low-input transcriptomic experiments often generate weak amplification signals, increased Cq variability and inconsistent detection of low-abundance transcripts. These issues are common when working with rare samples, single-cell material, FFPE-derived RNA, small biopsies or laser capture microdissection samples.

SpeAmp® supports qPCR preamplification by increasing the amount of selected cDNA targets before real-time PCR analysis, while keeping the workflow focused on selected genes, signaling pathways or biomarker panels.

1. Reverse transcription

RNA is converted into cDNA using an adapted reverse transcription workflow.

2. Specific preamplification

Selected cDNA targets are preamplified using SpeAmp® and appropriate pooled primers.

3. qPCR or qPCR array analysis

Preamplified cDNA is analyzed using qPCR assays, SignArrays® panels or selected gene expression workflows.

Preamplification performance depends on RNA quality, target abundance, reverse transcription efficiency, primer pool design, sample type and downstream qPCR conditions.
